What IVRT and IVPT each measure, how their methods differ and how to use them in microneedle development.

What does IVRT measure?

In-vitro release testing is used to compare release behavior from a formulation or microneedle structure. Medium, apparatus, sampling and analytical conditions must be chosen so the method can distinguish relevant formulation or process changes.

What does IVPT add?

In-vitro permeation testing uses skin and a diffusion system to assess the process of active entering or passing through skin. Skin source and integrity, application conditions, sampling and mass balance can materially affect the output.

When should each method be used?

IVRT is often useful for formulation and process comparisons; IVPT can address a skin-interface question. A product may need one, both or a different model depending on its mechanism and development objective.

What makes a method useful?

A useful method has a defined question, appropriate controls and evidence that it is sufficiently discriminating for its intended use. Regulatory requirements and validation expectations must be considered separately when applicable.
